Abstract

This research aims to determine the production technique for Gracilaria sp seaweed plantlets produced from spores, conducted in August - November 2024 at the Fish Reproduction and Genetics Laboratory, Pangkep State Polytechnic of Agriculture. Parameters that influenced spore growth were observed including light intensity, temperature and salinity of the culture medium. The research began by taking the thallus of Gracilaria sp. containing cystocarp and it was cut 1.5-2 cm long and soaked in sterile sea water with a salinity of 30 ppt. The thallus were maintained in a petri dish containing a culture medium PES (Provasoli's Enrich Seawater) fertilizer at a dose of 5 mL PES in 1000 mL seawater. Observation of the release of spores from the cystocarp was carried out 24 hours after planting. Observation of the growth of spores into plantlets was carried out at room temperature 26-27℃, using a microscope with 10-20X magnification. The results were that spore release occurred at DOC-2 and spore attachment occurred at DOC-9. The spore size increases up to DOC-14. Spore buds began to appear at DOC-18 and spore buds began to grow at DOC-22. The increase in the length of the spore shoots began at DOC-32 and the shoots began to produce branches at DOC-40. In DOC-52 the spore shoots have become plantlets. This research indicates that PES fertilizer can be a good medium for the growth of Gracilaria sp spores to become plantlets at a light intensity of 12 hours of light and 12 hours of darkness.
